Human Variability in Carboxylesterases and carboxylesterase-related Uncertainty Factors for Chemical Risk Assessment
نویسندگان
چکیده
Carboxylesterases (CES) are an important class of enzymes involved in the hydrolysis a range chemicals and show large inter-individual variability vitro. An extensive literature search was performed to identify vivo probe substrates for CES1 CES2 together with their protein content enzymatic activity. Human pharmacokinetic (PK) data on Cmax, clearance, AUC were extracted from 89 publications Bayesian meta-analysis using hierarchical model derive CES-related distributions related uncertainty factors (UF). The indicated that 97.5% healthy adults covered by kinetic default UF (3.16), except clopidogrel dabigatran etexilate. Clopidogrel is metabolised small amount polymorphic CYP2C19, which can have impact overall pharmacokinetics, while seen etexilate might be due differences absorption, since this influenced food intake. moderate high (<CV 50%), possible polymorphism enzyme but also sample size available per chemical. presented used combination vitro pathway-specific distributions.

Modeling of variability and uncertainty in human health risk assessment
Health risk assessments have been carried out worldwide to examine potential health risk due to exposure to toxic contaminants in various environments. In risk assessment, it is most important to know the nature of all available information, data or model parameters. It is observed that available information/data are tainted with uncertainty and variability in the same time, i.e., uncertainty a...
متن کاملShort Communication Characterization of Recombinant Human Carboxylesterases: Fluorescein Diacetate as a Probe Substrate for Human Carboxylesterase
Human carboxylesterase (CES) 1 and CES2 are members of the serine hydrolase superfamily, and both exhibit broad substrate specificity and are involved in xenobiotic and endobiotic metabolism. Although expression of CES1 and CES2 occurs in several organs, their expression in liver and small intestine is predominantly attributed to CES1 and CES2, respectively. We successfully expressed CES1 form ...

Human Health Risk Assessment of Cadmium Exposure in Dry Deposition of Atmospheric Dust (Uncertainty Analysis)
Background and purpose: The atmospheric suspended particulate matter is a major route of human exposure to different kinds of environmental pollutants especially toxic heavy metals. The aim of this study was to assess the carcinogenic and non-carcinogenic risk of cadmium in dry deposition of atmospheric dust in Qazvin, Iran. Materials and methods: Glass traps were used for sampling dry atmosp...
